| Application | 3-chloro-4-fluorophenyl magnesium bromide can be used as an intermediate in pharmaceutical synthesis, such as the preparation of pyrrolidine derivatives such as (2R,5S)-1-(tert-butoxycarbonyl)-5-(3-chloro-4-fluorophenyl) pyrrolidine-2-carboxylate, the compound has peptide calcitonin gene-related peptide (CGRP) receptor antagonism and can be used to treat various diseases associated with CGRP receptors. |
| Preparation method | Add trace iodine to the mixture of magnesium (434mg) and THF(2mL) under argon atmosphere. After the mixture is heated to 60°C, a solution of THF(6.5mL) of 4-bromo-2-chloro-1-fluorobenzene (3.58g) is added dropwise (the temperature is maintained at 65°C during dropwise addition). The mixture was stirred at 60°C for 30 minutes and then cooled to room temperature to obtain 3-chloro-4-fluorophenyl magnesium bromide. |
